Met behulp van onderzoekskaarten, bestaande uit 6 leugenkaarten en 6 waarheidskaarten, proberen spelers elkaar te misleiden. Om de beurt kiezen spelers een kaart en lezen ze hardop voor wat erop staat. Het doel is om je medespelers te overtuigen dat je de waarheid vertelt, zelfs als dat niet zo is. Als je vermoedt dat iemand liegt, kun je ze ontmaskeren door op de leugenaarsknop te drukken. Dit onthult een bewijskaart, waardoor spelers dichter bij de oplossing van de zaak komen. Het spel eindigt wanneer een speler de juiste persoon beschuldigt. Wie de juiste persoon beschuldigt, wint het Cluedo Leugenaars Spel!
